Based on years of clinical experience, the kit utilizes a double-antibody sandwich method to determine ALV-J expression in specimens. The microplate is pre-coated with purified antibodies that bind specifically to ALV-J antigens present in the sample. After washing away unbound substances, HRP-labeled secondary antibodies are added, forming an antibody-antigen-antibody complex. Following another thorough wash, TMB substrate is introduced. Under the catalytic action of HRP, TMB turns blue and then changes to yellow when an acid is added. The optical density (OD) at 450 nm is measured using a microplate reader, and results are compared against a cutoff value to determine whether ALV-J is present in the sample.
